Role of deoxyribonucleic acid polymerase beta in nuclear deoxyribonucleic acid synthesis.

چکیده
Exonuclease activity has been measured in preparations of both enzymes. The activity associated with DNA polymerase a requires denatured DNA, Mg2+ and is ATP-dependent. The polymerase b-associated nuclease requires denatured DNA and Mg2+ or Caz+. Studies of the distribution of these enzymes during the cell cycle have shown that extracts of cells harvested at the time of nuclear DNA synthesis (Chiang & Sueoka, 1967) have DNA polymerase b as the predominant DNA-synthesizing activity, whereas DNA polymerase a is the major activity in cells harvested at a resting phase of growth. The two major DNA polymerases of Chlamydomonas reinhardii therefore seem to differ in physical and enzyme properties and in the time of appearance during the vegetative cell cycle.

Template requirement of maize RNA polymerase.
Maize RNA polymerase utilizes heated deoxyribonucleic acid more effectively than native deoxyribonucleic acid as a template for ribonucleic acid synthesis. A ribonucleic acid-deoxyribonucleic acid hybrid accumulates in the presence of heated deoxyribonucleic acid. The amount of product formed with either native or heat-denatured deoxyribonucleic acid does not exceed the amount of deoxyribonucle...
متن کاملParticipation of deoxyribonucleic acid polymerase alpha in amplification of ribosomal deoxyribonucleic acid in Xenopus laevis.
Aphidicolin, a known inhibitor of eucaryotic deoxyribonucleic acid (DNA) polymerase alpha, efficiently inhibited amplification of ribosomal DNA during oogenesis in Xenopus laevis. DNA polymerase alpha, but not DNA polymerase gamma, as isolated from ovaries, was sensitive to aphidicolin. DNA polymerase beta was not detectable in Xenopus ovary extracts. Therefore, DNA polymerase alpha plays a maj...
متن کاملRole for deoxyribonucleic acid ligase in deoxyribonucleic acid polymerase i-dependent repair synthesis in toluene-treated escherichia coli.
In a toluene-treated mutant of Escherichia coli K-12 having a temperature-sensitive, conditionally lethal mutation in the structural gene for deoxyribonucleic acid (DNA) ligase, an extensive DNA repair synthesis occurred in X-irradiated cells at the nonpermissive temperature, 42 C. At the permissive temperature, 30 C, nearly normal semiconservative synthesis and limited repair synthesis were ob...
متن کاملEffect of drugs on deoxyribonucleic acid synthesis in isolated mammalian cell nuclei. Comparison with partially purified deoxyribonucleic acid polymerases.
In order to ascertain the identity of the DNA-dependent DNA polymerase responsible for the observed DNA synthesis in nuclei isolated from baby-hamster kidney (BHK-21/C13) cells a comparative study was carried out on the effects of some drugs, reported to influence DNA synthesis, on DNA synthesis catalysed by these nuclei and by partially purified DNA polymerase-alpha and -beta. In all cases DNA...
متن کاملEnzymatic synthesis of deoxyribonucleic acid. XII. A polymer of deoxyguanylate and deoxycytidylate.
The enzyme deoxyribonucleic acid polymerase, purified from Escherichia coli, catalyzes extensive formation of deoxyribonucleic acid by a mechanism that involves replication of a deoxyribonucleic acid primer. This replication requires the presence of the four deoxynucleoside triphosphates commonly found in deoxyribonucleic acid, and deoxyribonucleic acid itself (1). In the absence of one, two, o...
